Western blot: On transfected cells, use at a

transfected cells with antibody to CYLD (human).
Method: 10 μg per lane of cytosolic protein extract from
mock transfected HEK 293T cells (lanes 2, 4 and 6) or cells
transfected with a vector expressing FLAG-tagged human
CYLD (lanes 1, 3 and 5) were run on SDS-PAGE and
analyzed by Western blot. Lanes 1 and 2 were probed with
an anti-FLAG antibody. Lanes 3 to 6 were probed with
antibody to CYLD (human) at a 1:3,000 dilution. Detection
was completely abolished by the blocking peptide (preincubation
with 10 μM peptide for 1 hour) (lane 5). Detection
was carried out with an ECL system.

Method: HEK 293T cells transfected with a vector
expressing FLAG-tagged human CYLD were incubated with
antibody to CYLD (human) at a 1:200 dilution. An anti-rabbit
IgG antibody conjugated with FITC was used for detection.
Note: Detection with an anti-FLAG antibody gave an
identical pattern
